HMGB1是一種核蛋白，其由壞死細胞被動釋放，由凋亡細胞保留，并由炎癥細胞主動分泌（Dumitriu等人，HMGB1:從內部引導免疫。Trends Immunol 2005，26:381-7）。HMGB1對生命至關重要：HMGB1基因敲除小鼠出生后不久死亡。
HMGB1作為運動細胞的化學引誘劑和細胞因子的誘導劑的兩種不同活性被歸因于不同的生物化學形式。還原的HMGB1（具有化學引誘活性的形式）被還原；二硫化物HMGB1包含C23和C45之間的二硫鍵（完整注釋：HMGB1C23-C45C106h–Antoine J.等人（2014）。Mol-Med）。
二硫化物HMGB1可誘導單核細胞和其他炎癥細胞產生細胞因子和趨化因子（Venereau等人，2012）。
在結構上，二硫化物HMGB1在半胱氨酸殘基23和45與還原的半胱氨酸殘基106之間具有二硫鍵。測試二硫化物HMGB1刺激細胞因子的能力
